About the Founders: William Sellers is a core institute member of the Broad Institute of MIT and Harvard, a faculty member at Harvard Medical School, and faculty member and senior advisor to the president for experimental therapeutics at the Dana-Farber Cancer Institute. Eric Fischer is an Assistant Professor in the Department of Biological Chemistry and Molecular Pharmacology at Harvard Medical School and an Independent Investigator, Dana-Farber Cancer Institute. Responsibilities In this role, part of the Lead Discovery team at Civetta, you will work under the supervision of a Senior Scientist to implement a pipeline for expression and purification of human proteins in both bacterial and insect cell systems. Your skills in cell culture and protein purification will be critical to experiments designed to provide protein reagents for downstream work in high-throughput screening and structural biology at Civetta. This is an excellent opportunity to learn and grow in the unique environment of a start-up biotech in Cambridge. Key Responsibilities: * Perform small-scale and pilot-scale expression trials using robotic liquid handling systems to identify conditions for soluble expression of human proteins in heterologous systems * Scale-up to liter volumes of bacteria in high-density bio-fermentation systems, and of insect cells in shaking incubators * Learn and implement standardized purification workflows for multiple-column purifications of fusion-tagged proteins Qualifications Required Skills and Qualifications: * MS or BS degree in biochemistry, molecular/cell biology, or equivalent with 1-2 years of laboratory experience * Solid experience with standard molecular biology techniques including PCR, primer design, and subcloning into expression vectors preferred. * Exposure to protein biochemistry techniques in small scale, for example analysis of soluble expression using Western blotting and/or small-scale purification of protein utilizing affinity resins, is preferred. * Exposure to Opentrons or other robotic liquid handling systems, or willingness to learn these tools, is a strong plus. * Familiarity with standard gravity column purification procedures (i.e. nickel or GST affinity) and protein SDS-PAGE gel analysis desired.
